D3094: Do plants respond differentially to insect-mediated virus infection and insect feeding?

Infection of aphid transmitted poleroviruses could modulate gene expression associated with plant development and defense. It is unclear how gene expression profiles differ following aphid feeding and aphid-mediated transmission of a polerovirus between primary and alternate hosts. In this study, gene expression patterns following a polerovirus, cotton leafroll dwarf virus (CLRDV), infection in the primary host cotton and three alternate hosts viz., hibiscus, okra, and prickly sida were assessed. In addition, gene expression profiles were examined to tease out the differences between aphid feeding and aphid-mediated inoculation of CLRDV. For differential expression analysis, two treatment comparisons (CLRDV-infected vs. non-infected and mock-inoculated vs. non-infected) were evaluated for each host species. The CLRDV-infection identified 2079, 1238, 1484, and 1773 differentially expressed genes (DEGs) in cotton, hibiscus, okra, and prickly sida plants, respectively. The number of DEGs induced by aphid feeding was less than aphid mediated inoculation of CLRDV in all host species except okra. The functional annotation of DEGs using gene ontology (GO) and KEGG terms identified the genes associated with plant development and defense. The defense genes associated with salicylic acid, jasmonic acid, pathogen-related proteins, heat shock proteins, transcription factors and membrane transporters were differentially expressed between host species in CLRDV-infected plants. In summary, results indicated that the gene expression observed in CLRDV-infected plants varied substantially with host plants. Gene expression following aphid feeding also was not consistent across host species. Results indicated that differences in host susceptibility to the virus and/or the vector could differentially influence gene expression in hosts.
